- W2040418392 abstract "The changes in the amount, distribution, and reactivity with cytochrome b5 of the terminal enzyme of the hepatic microsomal stearyl CoA desaturation system, the cyanide-sensitive factor (CSF), were studied during the induction of desaturation activity by refeeding of fasted rats by analyzing the kinetics of reoxidation of microsomal bound cytochrome b5 in the presence of p-cresol. During the initial period of induction the reoxidation kinetics was biphasic, consisting of a rapid, p-cresol-stimulated phase and a slow phase which was not affected by the phenol. As the induction proceeded, the contribution of the rapidly reacting species was increased progressively. It was suggested that cytochrome b5 exists in the membranes both in a “functional” state where it is in interaction with CSF and in a “nonfunctional” state, and that the induction of the desaturation activity is accompanied by an increase in the percentage of “functional” cytochrome b5. In the rough microsomal membrane, the induction produced a continuous increase in the apparent firstorder rate constant (k′) of the p-cresol-stimulated reoxidation of cytochrome b5, but no clear biphasicity or linearity was observed during the initial period of induction. In the smooth membrane, on the other hand, both the percentage of “functional” cytochrome b5 and the k′ value of the p-cresol-stimulated reoxidation increased as the induction proceeded. These results suggested that newly synthesized CSF is first incorporated into the rough membrane to initiate an interaction with preexisting cytochrome b5 and then propagates to the smooth membrane. The microsomal membranes containing a high concentration of cytochrome P450 due to phenobarbital treatment could also accommodate CSF to constitute the desaturation system. It is concluded that CSF and probably also some other protein components of microsomes can be finally distributed homogeneously in the membranes, although a clear heterogeneity in the distribution of CSF is observable during the initial period of induction due to the fact that it is synthesized only on the rough membrane." @default.
