- W2040428215 abstract "We have studied the properties of mannosyl transferase from the mycelial and yeast forms of the dimorphic fungus Mucor rouxii. Most of the enzyme was found associated with the membrane fractions of the cells. Cell walls contained lower activities. Activity was maximal at pH 6.5 and 28°C and required a metal ion, Mn2+ and Fe3+ being the most effective ones. Proteases were either nonstimulatory or destructive of the mannosyl transferase activity. The exogenous acceptors mannan (Saccharomyces cerevisiae) and mucoran (M. rouxii) did not stimulate activity. Kinetics of the reactions were biphasic and two Km and Vmax values were calculated. Vmax values were similar for the enzymes from both mycelium and yeast, but Km values of the enzyme from the mycelial form were higher. Maximal activity of mannosyl transferase of the mycelial forms was obtained after 9 h of growth, whereas levels of the yeast enzyme started to rise after 12 h of growth. A shift of aerobic cultures to anaerobiosis produced a decay in the mannosyl transferase activity of the fungus, whereas transfer of an anaerobic culture to aerobiosis induced an increase in the levels of mannosyl transferase of the organism. These data are discussed in the light of a possible role of mannosyl transferase in growth and morphogenesis." @default.
