- W2040489768 abstract "The protonated form of denatured DNA is reduced in the adsorbed state in a totally irreversible electrode reaction to a strongly adsorbed product. At full coverage the amount of adsorbed DNA varies only slightly with the adsorption potential within a wide range, with the pH up to 7 and with temperature up to a threshold value. The protonated molecules of DNA are adsorbed via π-bonding of their pyrimidine and purine residues, while the other parts of the DNA molecules including the solvated sugar and phosphate group are orientated to the solution. In alkaline solutions no reduction of adsorbed denatured DNA is observed but still a reorientation in the adsorbed layer occurs at cetain electrode potentials. Native DNA is also adsorbed at the electrode and reduced in acid solution in the same totally irreversible electrode reaction as denatured DNA. However, for native DNA a further prerequisite for the electrode reaction is a conformational change in the adsorbed state under the influence of the electric field in the interfacial region leading to a certain extent of helix-coil-transition. The extent of the partial separation of both strands makes the electrochemically active sites of DNA accessible for the electrode reaction." @default.
