The Golgi apparatus was very heavily labeled 30 min after injection, and its radioactivity declined in the subsequent samples. A peak of radioactivity was observed 1–2 h after injection in groups of small vesicles in the cytoplasm. Labeling of the cell surface was initially low, but it then rose rapidly to a peak 12 h after administration of the precursor. Other types of cytoplasmic vacuoles, including large ‘fringed’ vacuoles, slowly increased in radioactivity to a maximum in the final sample prepared 24 h after injection. The results suggest that material is transported from the Golgi apparatus to the cell surface, and that the Golgi apparatus plays a role in the assembly of the cell surface coat. The pattern of labeling implicates aggregates of small vesicles and tubules (35 to 50 nm in diameter) in the transport of material from the Golgi apparatus to the surface. Other types of vacuoles, including the large fringed vacuoles (≈100nm to 1 μm in diameter) appear to be endocytosis vacuoles, since they become most heavily labeled after the peak labeling of the cell surface." @default.
