• W2041297285 abstract "Short segments of desheathed rat peripheral nerve were immersed in phosphate, tris-HCl and cacodylate buffers containing varying concentrations and admixtures of CaCl2, AgCl, MgCl2, PbCl2, and AlCl3 before fixation. These tissues were subsequently processed for electron microscopic examination. Axonal microtubules and neurofilaments were well-preserved following immersion in 0.1 M buffer, pH 6.5 to 7.5. Addition of 1 mM CaCl2 to the tris or cacodylate buffers resulted in a granular disruption of both neurotubules and neurofilaments. The disruptive effects of 5 mM CaCl2 could be prevented by chelation with equimolar concentrations of EDTA. A similar granular alteration of neurotubules and neurofilaments occurred after the addition of 10 mM AgCl, but not with MgCl2, PbCl2 or AlCl3 in concentrations up to 20 mM. Admixtures of MgCl2 did not prevent the calcium-induced disruption of these structures. The ultrastructural changes induced by calcium closely resemble the axoplasmic alterations encountered during Wallerian degeneration and suggest that excessive entry of calcium into the axonal compartment may play an important role in this phenomenon. The structural instability of neurotubules and neurofilaments in the presence of calcium also indicates the sensitivity of these organelles to calcium, a feature of possible consideration for the interaction of calcium in secretory mechanisms of cells." @default.
