This notion alone prevailed until the 1980s when it became evident that the heart secreted hormones that had effects on blood circulation. This profoundly changed our way of looking at the heart; it was not just a mechanical pump as previously thought but an endocrine organ as well. This conclusion was drawn when heart atrial extracts were injected into the blood circulation of a rat and a strong diuresis, natriuresis and vasodilation were observed. The exact physiological stimulus to which the heart atria responded by secreting hormones in physiological conditions was not known. At that time, an extensive review was published on another natriuretic hormone, a digitalis-like substance, which had been studied for more than 20 years [ [1] de Wardener W.E. Clarkson E.M. These findings led to the conclusion that the human body had both an endogenous diuretic and a digitalis acting on the cardiovascular system. In 1985 a seminal article was published in Nature in which Lang et al. [ [2] Lang R.E. Thölken H. Ganten D. et al. Atrial natriuretic factor: a circulating hormone stimulated by volume loading. Nature. 1985; 314: 264-266 Crossref PubMed Scopus (1041) Google Scholar ] described how a large and rapid intravascular volume increase produced high plasma levels of ANP in the rat blood circulation. This paper had a powerful effect on directing the studies with natriuretic peptides (both A- and B-type) towards the concept that a mechanical wall stress of the heart, either stretch or pressure, was under all conditions the key factor regulating the synthesis and release of natriuretic peptides. To date, over 25,000 articles have been published on natriuretic peptides and the majority of these studies have utilized the wall stress hypothesis in an effort to reveal the role of natriuretic peptides in various cardiac diseases, either as a diagnostic tool or as a guide of treatment.
