- W2041489734 abstract "Bone morphogenic protein -2 (BMP2; GeneID# 650) and interleukin-10 (IL-10; GeneID# 3586) genes have been implicated in bone and adipose tissue growth and maintenance. PURPOSE: We hypothesized that subjects with the homozygous rare alleles for BMP2 rs 15705 SNP would have significantly more adipose tissue and greater bone density compared to those with the homozygous common alleles, and subjects with the homozygous rare alleles for IL-10 rs1800872 SNP would have less adipose tissue and greater bone density compared to those with the homozygous common alleles. METHODS: Genotyping was performed using a novel TaqMan allele discrimination. Body composition was assessed using dual energy x-ray absorptiometry (DEXA) that provided total bone mineral density (BMD), fat free mass (FFM), and % fat mass (%FM). Analysis was conducted using an ANCOVA, and a post-hoc Sidak-Dunn was used to detect specific significant difference. RESULTS: 136 subjects were tested, and all genotype groups for BMP2 and IL-10 were in Hardy-Weinberg equilibrium. There was a trend for association (p=0.0507) of total body %FM and BMP2 genotypes in women; women homozygous for the rare allele CC had greater total body %FM (35.3 ± 1.9%) compared to women homozygous for the common allele AA (32.8± 0.9%). The association of FFM and BMP2 genotypes in women was significant (p=0.02); women heterozygous for the rare allele AC had less FFM (37.5 ± 0.6 kg) compared to women homozygous for the common allele AA (39.7± 0.5 kg). There were no significant associations of body composition with BMP2 genotypes in men. However, the association of total body %FM and IL-10 genotypes in men was significant (p=0.02); men homozygous for the rare allele AA had less total body %FM (13.6 ± 3.7%) compared to men homozygous for the common allele CC (23.9± 1.0%). There were no significant associations of body composition with IL-10 genotypes in women. CONCLUSIONS: These data indicate BMP2 genotype is associated with body composition phenotypes in women, and IL-10 genotype is associated with body composition phenotypes in men. Identifying genes associated with body composition may assist in producing a screening tool to determine an individual's risk for diseases (e.g. Type 2 Diabetes) associated with body composition.
