- W2042012487 abstract "The inhibitory action of starved rat serum on the in vitro metabolism of cartilage from hypophysectomized rats was examined. Serum was obtained from pituitary-intact rats (90–100 g BW in a fed condition) which had been fasted for 4 days. The effects on sulfate incorporation into proteoglycans were generally used as a measure of inhibition, although thymidine incorporation into DNA was also impaired by starved rat serum. Such serum had sufficient inhibitor to overcome the somatomedin activity of an equivalent volume of normal rat serum. The dose-response slope for somatomedin activity was lowered progressively by increasing concentrations of inhibitory serum added to normal rat serum, but a reduction of somatomedin activity by more than 90% was required for statistically significant evidence of nonparallelism. The striking antagonism of the somatomedin activity of normal rat serum provided the basis for a quantitative assay of somatomedin inhibitor. Increments of starved rat serum in a medium with a fixed concentration of normal rat serum caused a more precipitous fall of somatomedin activity than a proportional dilution of normal rat serum alone. The action of somatomedin was limited more effectively than that of insulin at concentrations which produced comparable effects on cartilage. Therefore, the inhibition cannot be attributed to a simple toxic effect. The pituitary is not essential for somatomedin inhibitory activity, since serum from fed hypophysectomized rats was about half as potent as starved rat serum. The somatomedin inhibitory activity of starved rat serum was labile at 60 C, but inactivation at that temperature was reduced by lowering the pH and diluting the serum before heating. The serum with minimal dilution at pH 2.0 could be heated at 60 C for 1 h without decreasing inhibitory activity. Dilute serum at pH 5.5 lost inhibitory activity on heating at 100 C for 10 min. However, a further lowering of the pH to 2.0–3.5 permitted heating at 100 C with retention of most of the inhibitory property in solution. Somatomedin inhibitory activity was unaffected by repeated freezing and thawing, resistant to destruction at pH 11.0 for 3 h at 37 C, precipitable with acetone from serum containing n-butanol, and extractable into acid-ethanol. Most of the activity was associated with acidic protein fractions prepared by ion exchange chromatography, and the peak of activity on gel filtration at pH 7.4 was present in fractions that were eluted in a range corresponding to an apparent molecular size of 27,000–40,000." @default.
