- W2042171502 abstract "1820 The capillary supply is reduced in obese muscle. Vascular endothelial growth factor (VEGF) is an important angiogenic growth factor for the maintenance and expansion of the skeletal muscle capillary supply. PURPOSE: We hypothesized that, compared to lean (LN) subjects, obese (OB) subjects would demonstrate reduced VEGF and VEGF receptor expression at rest and in response to acute exercise. METHODS: Eight LN (BMI = 22.4 ± 1.1 kg/m2) and 8 OB males (BMI = 36.1 ± 1.8 kg/m2) exercised for 1 hr at 50% of VO2max. Muscle biopsies were obtained from the vastus lateralis prior to and at 2 hr after the completion of the exercise bout. VEGF, KDR, and Flt-1 mRNA and protein were analyzed by Northern blot and ELISA technique, respectively. Muscle morphology was performed by standard techniques. RESULTS: Exercise significantly increased VEGF, KDR, and Flt-1 mRNA. There were no differences in mRNA or protein at rest or exercise between groups. Capillary density and capillary-to-fiber area ratio were lower in OB. CONCLUSIONS: The lower skeletal muscle capillary supply observed in obese muscle does not appear to result from reduced VEGF or VEGF receptor expression.
