- W2044135302 abstract "RATIONALE: IgE antibody assay accuracy is important since levels are sometimes used to predict clinically-relevant food allergy and confirm allergen sensitization. We hypothesized that allergen-specific IgG antibody interferes in the accuracy of IgE antibody serological assays using limited allergen (chip-based systems). METHODS: Sera containing varying amounts of IgE and IgG anti-casein were pre-adsorbed with Protein-G Sepharose (HiTrap, GE Healthcare) to remove IgG or uncoupled Sepharose (sham control). Specimens were then analyzed by ImmunoCAP and ISAC (Phadia-VBC Genomics) to confirm removal of IgG antibody and examine IgE antibody level changes following IgG removal. RESULTS: The Protein-G Sepharose absorption removed 97.7%+/-3.3% of IgG anti-casein. ImmunoCAP measurement of IgE anti-Casein was resistant to IgG anti-Casein interference (116% +/- 20% change) even with IgG/IgE molar ratios >200,000. In contrast, the chip-based ISAC (using more limited casein-allergen binding sites) displayed IgG anti-casein dose-dependent increases up to 991% in the quantity of detected IgE anti-casein following removal of IgG anti-casein. CONCLUSIONS: Interpretation of IgE antibody results from chip-based assays using limited allergen (e.g., ISAC or epitope mapping studies using peptides on chips) should be viewed cautiously, especially when testing sera from patients on immunotherapy. Negative results may falsely occur due to IgG antibody interference. IgG removal prior to chip assay testing may be necessary to maximize accuracy." @default.
