- W2045039106 abstract "腎不全時には，水分・電解質バランスの異常や老廃物の蓄積により生命が脅かされることから，透析療法が導入されることも少なくない．わが国の透析導入患者数は増加の一途をたどる現状であり，腎疾患治療薬の更なる開発が望まれる．著者らは，急性および慢性腎不全モデル動物を作製し，その発症・進展機構とそれらに有効な治療薬について研究している．従来から急性腎不全モデルとしては，虚血再灌流，重金属，各種薬物などによる腎機能低下モデルが用いられているが，主に著者らは腎臓の血流を一時的に遮断した後，その血流を再開通させることで発症する腎虚血再灌流障害モデルを用いている．技術的にも比較的容易であることから，安定した腎機能障害動物が得られ，実験者間の個人差も比較的少ない．費用の面でもきわめてリーズナブルである．本モデルを用いて著者らの研究室では，腎虚血再灌流障害の発症と進展に関わる種々の因子を同定するとともに，その障害をきわめて効果的に改善する薬物も見出している．一方の慢性腎不全モデルでは，腎部分切除や腎動脈分枝を結紮することにより，機能糸球体数を物理的に減少させて慢性的に腎障害を引き起こす方法が用いられることが多い．また最近では，糖尿病誘発性のモデルを用いた例も多くみられる．本稿では，誌面の都合上，急性腎不全モデルとして腎虚血再灌流障害，慢性腎不全モデルとして腎部分摘除の各モデルを取り上げ，動物の作製方法について解説する．さらに，腎機能低下の程度や進行並びに組織病変はそれぞれの病態モデルで特徴的であるため，それらがわかるように著者らの実験結果を例に挙げて記述する．" @default.
