- W2046163056 abstract "Magnesium, acting from the serosal surface, depressed or abolished contractions of guinea-pig isolated ileum caused by acetylcholine, 5-hydroxytryptamine, histamine and nicotine added to the organ bath. A parallel shift in dose-response curves was obtained with the lower concentrations of magnesium. 5-Hydroxytryptamine and nicotine were about twice as sensitive to the blocking effect of magnesium than was acetylcholine or histamine. In another series of experiments, magnesium, acting from the serosal or from the mucosal surface, depressed or abolished the peristaltic reflex of guinea-pig isolated ileum. Concentrations of magnesium, which had an approximately equal blocking effect on the reflex, were about 10 times higher when administered into the lumen of ileum. Choline esters (acetylcholine, methacholine, propionylcholine and butyrylcholine), anticholinesterases (eserine and neostigmine), ganglionic stimulant agents (nicotine and dimethylphenylpiperazinium) and polypeptides (angiotensin, eledoisin and bradykinin) acting from the serosal surface, antagonized the peristaltic block caused by magnesium added to the organ bath. 5-Hydroxytryptamine, histamine, pilocarpine and arecoline acting from the serosal surface did not remove the peristaltic block. Methacholine and eledoisin, applied to the mucosal surface, restored the peristaltic waves previously abolished by intraluminal administration of magnesium but acetylcholine, 5-hydroxytryptamine, histamine and bradykinin administered intraluminally were ineffective. It is suggested that the predominant action of magnesium on the peristaltic reflex takes place at the cholinoceptive sites, which ensure peristaltic waves, in the ganglia of myenteric plexus. In part, magnesium blocked the peristaltic reflex by an action on the nerve-muscle junction in the longitudinal muscle. However, the antiacetylcholine effect of magnesium is not specific and probably is due in part to a “stabilizing” effect of the cation on membranes." @default.
