The induction of rat liver glucokinase by glucose was investigated under conditions where (1) the secretion of insulin is blocked and (2) the normal utilization of glucose by the liver is prevented. 2. The induction of hexokinase II of adipose tissue by insulin in vitro was studied in the absence of glucose, with pyruvate as a substrate. 3. A double-labelling technique was used to measure directly the incorporation of amino acids into each enzyme molecule. 4. Glucokinase and hexokinase II activity were studied in hypophysectomized and thyriodectomized rats and after chronic administration of thyroxine and hydrocortisone to such animals. Both these hormones had dramatic effects on the enzyme activities. 5. It will be shown that insulin is necessary for the adaptive formation of both glucokinase of liver and hexokinase II of adipose tissue; that insulin has a role in addition to the of favoring glucose entry; that the hormone's effect involves de novo enzyme formation; and that the insulin-stimulated process involves intermediates of different half-lifes in liver and adipose tissue and whose formation is suppressed by actinomycin D. 6. The hypothesis will be put forth that the actual role of the hormone is indirect, being an effect on levels of metabolites." @default.
