- W2046966427 abstract "Introduction: Homeostasis between indigenous intestinal flora and host immune responses may be broken in inflammatory bowel disease. Our study tests the hypothesis whether repeated preventive oral administration of live probiotic bacterial strains Escherichia coli O83:K24:H31, Escherichia coli Nissle and Lactobacillus casei immunitas (LCI) could protect mice against dextran sodium sulphate (DSS) induced colitis. Methods: E.coli and Lactobacillus casei immunitas were cultivated and isolated in stationary growth phase. They were washed 3 times and diluted in PBS (Phosphate Buffered Saline) to final concentration 10 9/ml. BALB/c mice (n=75) has been fed daily for 14 days by gastric gavage with 1001/4l of live bacterial suspension or with 1001/4l of PBS in control group. DSS colitis was induced by administering 5%DSS in drinking water. The severity of colitis was evaluated one week later. The presence of rectal prolapse, rectal and colonic bleeding and weight loss were recorded. A symptom score was constructed. The lenght of the colon was measured and colonic samples were obtained for histopathology. For immunological evaluation serum and enteral content were collected and antibody titers were determined by ELISA. Results: We observed decrease in symptom score in the mice fed with probiotic strains compared with PBS fed control animals. While severe inflammation, crypt destruction and ulceration of the mucosa was observed in the PBS-fed group by histology evaluation, less inflammation was observed in the LCI-fed group. Mice exposed to E. coli suspension had an inflammation histology score comparable to PBS controls. Mice fed with water instead of DSS had no colonic inflammation regardless of probiotic strain they had previously received indicating that the administration of tested probiotic strains had no intrinsic harmful effect on the colonic mucosa. Application of LCI stimulated IgA antibody production in the gut and systemic IgM and IgG antibody production when compared to non-treated mice. Conclusion: Our data extend the evidence that non-pathogenic luminal bacteria participate in the pathogenesis of chronic intestinal inflammation. Administration of probiotic bacteria influencing the composition of intestinal flora may be relevant to the development of novel strategies in the prevention and management of IBD patients.
