• W2047701116 abstract "Abstract The present investigations were initiated to examine the ultrastructural aspects of intestinal fat transport and chylomicron formation in rats treated with inhibitors of protein synthesis. Female rats were injected intraperitoneally with puromycin, ethionine or cycloheximide. Fatty chyme was injected into ligated segments of jejunum after suitable periods. With the first two inhibitors, additional segments were injected with radioactive leucine. Following 30 minutes absorption, the chyme-injected segments were prepared for electron microscopy. Labeled segments were assayed for synthesis of radioactive protein. A significant decrease in protein synthesis was shown in rats treated with puromyein or ethionine. Examination of absorptive cells from these treated animals did not revcal evidence for cellular damage or an abnormal pattern of fat transport. Compared with control animals, a similar intracellular distribution of lipids was found. Furthermore, chylomicrons were consistently present in large numbers in extracellular spaces of the epithelium and lamina propria. Similar results were obtained with cycloheximide; however, evidence for cellular alterations were observed in some cells. The results suggest that the relation of protein synthesis to chylomicron transport from the intestine requires further study." @default.
