- W2048255415 abstract "RATIONALE: Some patients with walnut food allergy report allergy to other nuts, including macadamia. We sought to clarify whether this could be due to IgE cross-reactivity between walnut and macadamia nut. METHODS: Sera were obtained from three patients with reported macadamia and walnut allergy (one proven by food challenge to both, two with severe reactions to tree nut ingestion and excellent histories of clinical reactivity to both macadamia nut and walnut). IgE inhibition immunoblotting to macadamia nut extract separated by 15% SDS-PAGE and transferred to nitrocellulose was performed by pre-incubating sera with extracts of macadamia, walnut, cashew, ryegrass pollen, rJug r 1 (recombinant walnut 2S albumin), rJug r 2 (walnut vicilin-like protein), or rJug r 4 (walnut legumin-group protein). RESULTS: IgE from all three serum samples bound multiple polypeptides, including a band at approximately 15 kDa that was not absorbed out by pre-incubation with grass pollen extract. Pre-incubation with walnut extract, but not cashew, was able to inhibit binding to this polypeptide in all samples. rJug r 1 pre-incubation also inhibited binding to the polypeptide in two serum samples, but no effect was seen with rJug r 2 pre-incubation on any IgE reactive band. rJug r 4 inhibited IgE binding to a polypeptide at approximately 19-20 kDa in two of the samples. CONCLUSIONS: Macadamia nut appears to contain walnut cross-reactive IgE binding proteins that are likely 2S albumins and legumin-group seed storage proteins based on the ability of rJug r 1 and rJug r 4 to inhibit IgE binding." @default.
