- W2048301903 abstract "Recent evidence indicates that cerebral ischemia (CI) and pathological factor of Alzheimer's disease (AD) such as β-amyloid(Aβ) protein can interact with each other, and even leads to exacerbated cognitive impairment. T his study examined cerebral blood flow (CBF) and blood volume (CBV) for 4 weeks in rats b y using dynamic contrast-enhanced computed tomography (DCE-CT or CT perfusion). The hypothesis is that Aβ and CI together may cause elevated hemodynamic dysfunction because cerebral hemodynamics is one of the first physiological markers affected by this combined interaction in the brain. The objective is to study the interaction between CI and Aβ in causing more severe hemodynamic disturbance in the early pathological stage using DCE-CT. For methods, AD was induced by bilateral injection of Aβ 25-35 peptides into cerebroventricles, and CI by unilateral injection of a vasoconstrictor, endothelin-1 into the right striatum of male Wistar rats. The combination (AD+CI) group included the above two types of injection. Image registration was done based on digitized stereotaxic atlas of rat brain anatomy. Specific region of interest (ROI) was drawn on the registered images for CBF and CBV analysis. Histopathological examination was performed for each rat to investigate neurodegeneration and inflammation. For results, DCE-CT maps revealed that ipsilateral striatum of AD+CI group showed relatively lower CBF and CBV than in AD alone and control rats within the acute phase and 1st week. In addition, AD+CI group resulted in a longer-lasting hemodynamic dysfunction than AD alone and control within 4 weeks. Histopathology currently is still undergoing and preliminary results showed consistency with imaging evidence so far. This CT perfusion study can quantitatively monitor the hemodynamic disturbance on parametric images in the rat model of combined AD pathology and cerebral ischemia." @default.
