- W2048392563 abstract "Abstract The thermotropic phase behaviour of mixed aqueous dispersions of dipalmitoylphosphatidylcholine (DPPC) and 1,2-dilauroylglycerol (DLG) has been characterized by differential scanning calorimetry, synchrotron X-ray diffraction and freeze-fracture electron microscopy. The equimolar mixture of DPPC and DLG showed a melting transition at Tm=53°C from a lamellar gel to a hexagonal fluid phase which is approx. 10°C above the melting points of both pure components. This indicates the formation of a strong 1:1 compound of DPPC and DLG. Mixtures with low amounts of DLG up to 10 mol% showed a thermotropic phase behaviour similar to the pure phospholipid: heating from 4 to 60°C revealed a Lβ–Pβ′–Lα phase transition sequence in the small-angle X-ray region. The simultaneously recorded differential scanning calorimetry (DSC) signal showed only slight variations in the width and positions of the endothermic transitions. Freeze-fracture electron microscopy showed a gradual increase in the periodicity of the ripples at 40°C from 14 nm for pure DPPC to approx. 30 nm for 10 mol% DLG. Significant differences were observed for the samples containing 15 and 20 mol% DLG. The SAXS patterns showed a single lamellar phase undergoing a direct transition from a gel state to the fluid state. The DSC signal, however, clearly showed two melting events, indicating gel phase immiscibility. Freeze-fracture replicas showed that in the gel phase prior to chain melting the bilayers had either a smooth or a stripe-like appearance. We suggest, that dilauroylglycerol is miscible with DPPC in the gel state up to 10 mol%. When more than 10 mol% of DLG is present phase separation of a DLG-poor phase (DPPC:DLG, 9:1) and a DLG-rich phase (the stable 1:1 compound) occurs.
