- W2048708462 abstract "ISEE-371 Introduction: Spacial distribution analyses indicate six regions in the country with excess mortality over the national mean value. Within the framework of a multicenter ongoing study, the aim was to investigate the interaction between exposures to various sources and potential genetic susceptibility factors in two of the specific regions. Methods: Two counties exhibiting excess lung cancer mortality rate have been selected. The study population comprised a total of 217 primary lung cancer cases and 180 hospital-based non-cancer matched controls. The major histological types of lung cancer were squamous cell carcinoma (40.5%), adenocarcinoma (32.2%) and small cell carcinoma (15.7%). Detailed information on smoking, lifestyle, diet, occupational and environmental exposure was obtained with informed consent from the subjects by trained interviewers. GSTM1, GSTT1 and GSTP1 Ile105Val genetic polymorphisms of glutathione S-transferases have been determined from DNA isolated from peripheral blood samples by PCR-based methods. Statistical analyses were performed by STATA 7.0 software. Results: Smoking was the major risk factor for squamous cell carcinoma and small cell carcinoma. None of the three GST polymorphisms alone exhibited significantly increased risk for lung cancer. After stratification for smoking status and sex, the most pronounced effects of the GST genotypes were observed for ever-smoking male patients. GSTM1 homozygous deletion genotype slightly increased the risk for squamous cell carcinoma [odds ratio (OD) 1.40, 95% confidence interval (Cl) 0.77-2.53]. On the contrary, GSTM1 null genotype appeared to reduce risk for small cell carcinoma (OD 0.26, Cl 0.080-0.84, p=0.021). The effect of GSTT1 homozygous deletion was opposite for small cell carcinoma (OD 1.56, Cl 0.50-4.83) and adenocarcinoma (OD 0.41, Cl 0.15-1.16). The GSTP1 Ile/Ile genotype had the most pronounced association with adenocarcinoma (OD 1.94, Cl 0.99-3.80). Conclusions: The results suggest, that GST genetic polymorphisms may have special impact on the development of different histopathological types of lung cancer. In-depth ambient environmental and occupational exposure analyses in combination with multiple genotypes are expected to provide better insight of the causal factors of spacial differences of lung cancer mortality in the country." @default.
