- W2049012034 abstract "This communication reports the results of the examination in the electron microscope of the cylindrical particles of two strains of tobacco rattle virus. Negative staining was employed and certain of the pictures obtained were examined by the technique of optical diffraction. It was found that the uranyl formate stain reveals considerably more of the substructure of the virus than any other reagent tried. The basic helix of the particle is clearly visible, and the principal higher helices can also be seen occasionally. Optical diffraction shows that the apparent hand of the basic helix is predominantly left. Where they can be observed at all, features of the optical diffraction patterns due to the higher helices show that there are 3q+1 subunits in three turns (where q is an integer), as is the case with tobacco mosaic virus. No reason was found to dispute the value of q (25) suggested by the work of others on repolymerized tobacco rattle virus protein. The dimensions of the particle found here were similar to those reported by other workers. The diameter was 256 ± 3 A and the pitch 25·5 ± 0·3 A. In addition, an annular feature was observed at a radius of 82 ± 4 A; it cannot be said whether or not this indicates the position of the RNA. Unlike tobacco mosaic virus, particles in the “stacked disc” configuration were rarely seen, even in old samples. A corresponding difficulty was experienced in repolymerizing isolated protein to rods of any great length. Reconstitution of protein and RNA to form short lengths of rod is extremely simple. From all these measurements, the molecular weight of the subunit can be shown to be about 24,000, in agreement with chemical analyses. Once the length is known, it is possible to calculate the molecular weight of the protein in the completed particles. In the long type of particles this value is about 44,000,000 (1830 molecules), and for the short type about 16,000,000 (660 molecules). There may be four nucleotides per protein molecule, in which case the completed particles have molecular weights of 46,000,000 and 16,500,000, respectively." @default.
