- W2049284966 abstract "OBJECTIVE: A combination of factors contribute to oocyte quality including, aneuploidy and gene products essential for fertilization and embryonic development. Oocyte quality is a primary contributor to human reproductive failure and has been strongly associated with advanced reproductive age. However, oocyte quality may also play a role in cases of women who present with unexplained infertility and are not considered of advanced reproductive age. This study investigated the protein profiles of MII oocytes from young and mid reproductive age female mice. DESIGN: Prospective laboratory study using a murine model. MATERIALS AND METHODS: Metaphase II (MII) oocytes were collected from BDF-1 female mice of the following age groups: A) 6 weeks old; B) 20 weeks old. All female mice were primed with 5 IU hCG 13 hrs prior to oocyte collection. MII oocytes were isolated and processed in groups of five (n=16 replicates per group), prior to analysis by surface enhanced laser desorption ionization time-of-flight mass spectrometry. RESULTS: Oocyte maturity was recorded and no significant differences were observed between oocytes considered of young reproductive age and mid reproductive age. Oocyte protein profiles were captured towards the low molecular weight range (<30kDa) and were normalized to total ion current intensity prior to statistical analysis. The Mann-Whitney non-parametric test revealed a set of 4 proteins that were differentially expressed between the two groups: 3068 Da, 3619 Da, 4154 Da and 18230 Da (P<0.01). Identification studies are underway to characterize these differential expressed proteins according to reproductive age and potentially define their involvement during development. CONCLUSIONS: Initial data indicates differential protein expression between young and mid reproductive age mouse MII oocytes. Further studies to characterize the molecular signature of MII oocytes may benefit women who present for infertility treatment with unexplained prognosis and are not considered of advanced reproductive age." @default.
