- W2049673959 abstract "Studies were undertaken to understand the control of synthesis, stability and modification of UDP galactose epimerase and DNA-dependent RNA polymerase during sporulation of Saccharomyces cerevisiae. When a pre-induced culture of an inducible strain (wild type) is transferred to sporulation medium, the epimerase is inactivated to an undetectable level within 16 hours. Surprisingly, the addition of cycloheximide, a protein synthesis inhibitor, during sporulation stabilizes the epimerase activity. However, in a constitutive strain, the epimerase continues to be synthesized de novo during sporulation. Since the enzyme is synthesized during both vegetative growth and sporulation constitutively, the controls for synthesis of epimerase must be similar under these physiologically different conditions. After chromatography on DEAE Sephadex, there is no change observed in the elution patterns of RNA polymerase forms extracted from acetate growth vegetative cells, sporulating cells or from mature asci; in all cases RNA polymerase consists of three forms, Ib, II and III. However, single spore suspension obtained from asci by treatment with zymolase contains a new form with chromatographic properties similar to those of form Ia. Our data suggests that form Ia may be a modification product of form Ib. Le but de ce travail est d'élucider les contrôles de la synthèse de la stabilité et la modification de l'épimérase de l'UDP galactose d'une part, et d'autre part des RNA polymérases DNA-dépendantes pendant la sporulation de Saccharomyces cerevisiae. Lorsqu'une culture pré-induite d'une souche inductible (type sauvage) est transférée dans le milieu de sporulation, l'activité de l'épimérase décroît; elle est indécelable après 16 heures. L'addition de cycloheximide, inhibiteur de la synthèse des protéines, pendant la sporulation stabilise l'activité de l'épimérase. Cependant, dans une souche constitutive, l'épimérase est synthétisée de novo, tout au long de la sporulation. Puisque l'enzyme est synthétisée dans une souche constitutive à la fois pendant la croissance et la sporulation, les contrôles de la synthèse de l'épimérase doivent être similaires dans ces conditions physiologiques différentes. Aucune modification n'a été observée après chromatographie sur DEAE-sephadex dans les profils d'élutions des RNA polymérases extraites soit de cellules sporulantes, soit de spores. Dans tous les cas on trouve trois formes de la RNA polymérase: Ib, II III. Toutefois, les spores individuelles obtenues en traitant les asques avec de la zymolase présentent une nouvelle forme de l'enzyme dont les propriétés chromatographiques sont similaires à celles de la forme Ia. Nos résultats suggèrent que la forme Ia est une modification de la forme Ib." @default.
