- W2050871071 abstract "Abstract One group of young guinea pigs was anesthetized and fixed by perfusion with glutaraldehyde. Some epiphyses were post-fixed with OsO 4 and the sections stained with heavy metals. Other epiphyses were embedded without post-fixation and the sections were not stained. A third group of epiphyses were shockfrozen, freeze-dried and embedded without staining. This group was most suitable for crystallographic analysis. Contact of the latter ultrathin sections with water was less than 30 seconds. On the average the first mineralized clusters are larger than the unmineralized matrix vesicles. This led to the conclusion that already at an early stage of mineralization intra- and extra-vesicular regions are involved. This assumption is supported by the observation of a round or eliptic line of crystals which encircle compartments within the mineralized clusters. The diameters of the compartments correspond with those of the matrix vesicles and the lining crystallites seem to represent the mineralization of certain sites at the vesicle membrane. The lateral distances between the centers of neighboring mineralized clusters and those of neighboring matrix vesicles lie in the same range of 0.2.–0.8 μm but the distances between the vesicles are on the average somewhat shorter. These findings suggest that the formation of the mineralized clusters can be explained by nucleation within or on matrix vesicles. On the average, the lateral distances between the centers of parallel apatitic needles in these clusters are statistically significantly shorter than those of the crystals in collagen.
