Accordingly, catalytic turnover, by virtually all lipolytic enzymes, occurs at the interface of organized aggregates formed in aqueous dispersions of amphipathic and “lipidic” substrates. This chapter discusses the kinetics of interfacial catalysis by a water-soluble lipolytic enzyme that must attach to the substrate aggregate as a prerequisite for catalytic turnover. It focuses on a class of 14-kDa secreted phospholipases A2 (PLA2) that catalyze the hydrolysis of the sn-2-acyl chain of naturally occurring phospholipids to form lysophospholipids and fatty acids. In describing the kinetic behavior of PLA2, the emphasis here is on the formulation of experimentally testable questions and on the design of strategies and protocols necessary to resolve the primary rate and equilibrium parameters. Because only a few interfacial enzymes have been adequately characterized, the formalism developed here should be considered as a prototype to build a general conceptual understanding of the kinetic and molecular aspects of interfacial catalysis by membrane-bound proteins." @default.
