- W2051286201 abstract "Paraoxonase (PON) is an esterase which has an ability to reduce oxidization of low-density lipoprotein, hence play an important role in atherosclerosis. However, there have no studies evaluating whether polymorphisms of genes encoding PON family has any effect on systemic oxidative stress. In the present study, we investigated the association between gene polymorphism encoding PON1 and PON2 and systemic oxidative stress condition. Two-hundred thirty one community residents aged over 50 participated in the study. Informed consent for the procedure was obtained from each subject. Five SNPs of PON1 gene (−907C/G, −162G/A, −107C/T, Leu55Met, Gln192Arg) and one SNP of PON2 gene (Cys311Ser) were determined. As a systemic oxidative stress, urinary levels of PGF2alfa, which is a non-enzymatic product by oxidative stress from arachidonic acid, were measured by EIA. We observed linkage disequilibrium between PON1 −907C/G and −107C/T, and between −162G/A and Leu55Met. Urinary 8-epi PGF2alfa was significantly higher in smokers than non-smokers (433.7±207.2 vs 290.5±134.9 pg/mg creatinine, p<0.001). There was no significant association between urinary 8-epi PGF2alfa and any SNPs studied in the study in the total population. However, smokers with PON1 −107TT genotype had significantly higher urinary 8-epi PGF2alfa than smokers with CC+CT genotypes (ANCOVA:F[1,227]=8.44, p=0.004). General linear model analysis indicates interaction between smoking and PON1-107C/T genotype was an independent predictor for urinary 8-epi PGF2alfa. These findings indicate that −107TT genotype in the promoter regions of PON1 gene is a significant risk factor for oxidative stress in smokers.
