- W2051421321 abstract "To determine the biological effectiveness of 125I radioactive seeds continuous low-dose-rate irradiation on the human esophageal cancer cell line KYSE150 in vitro and explore cellular mechanisms. The KYSE150 cells were exposed to radiation of 160KV X-ray at a high-dose-rate of 1.052Gy/min and 125I radioactive seeds at a low-dose-rate of 2.77 cGy/h. Two groups were employed: single dose radiation group (SDR) and 125I radioactive seeds continuous low-dose-rate radiation group (125I-CLDR). Specific dose (4Gy) was chosen after preliminary experiments (data does not show) and cells without irradiation as the control. The responses of KYSE150 cells to two modes of radiation were evaluated by the colony-forming assay. MTS proliferation assay, cell cycle progression as well as apoptosis analysis. Furthermore, we detected the expression patterns of γH2AX, Bim and caspase-3 by Western blotting. The relative biological effect for 125I seeds compared with 160KV X-ray was 1.56. 72hrs after 4Gy irradiation, the inhibition effects showed significant difference between SDR and 125I-CLDR(t=11.92. P=0.001). The difference between proportions of cells at G2/M phase of SDR and CLDR groups was statistically significant(t=6.68, P=0.003), so as the apoptosis rate(t=16.29, P<0.0001). Moreover, γH2AX, Bim and caspase-3 expression levels in 125I-CLDR group increased compared with SDR groups. Compared with the X-ray high-dose-rate irradiation, 125I radioactive seeds continuous low-dose-rate radiation showed more effective proliferation inhibition, stronger induction of cell apoptosis and G2/M cell cycle arrest. Furthermore, 125I radioactive seeds continuous low-dose-rate radiation could strengthen DNA damage and apoptosis level, which indicated by up-regulating γH2AX,Bim and caspase-3 expression." @default.
