- W2051668459 abstract "Abstract 1. 1. Rings of pig aorta were digested with several proteolytic enzymes before submission to tensile strength measurements and light microscopy. The same procedures were also used on rings of isolated aortic elastin, the residues of formic acid digestion. 2. 2. The tensile strength of intact aorta was weakened in the decreasing order: papain and pepsin (100%) > pronase, elastase and collagenase (90%)>chymotrypsin (65%)>trypsin (50% loss of strength). 3. 3. The effect on isolated aortic elastin was uniformly high, with the exception of collagenase and trypsin which produced no significant weakening. 4. 4. Microscopically only collagenase produced apparent lysis of the collagen moiety, but elastase, pronase and papain all appeared to lyse elastin. Although tinctorically normal, both collagen and elastin in most instances were affected by the enzymes, as judged by tensile strength measurements. 5. 5. The persistence of normal staining collagen with reduced tensile strength is explained on the basis of enzymatic excision of NH2-terminal telopeptide regions on which cross-linking and tensile properties depend, and an absence of effect on the collagen helix, which provides the stainable fiber. 6. 6. Additional studies revealed significant weakening of aorta by non-proteolytic bacterial α-amylase, which suggests a role for glycoproteins in tensile strength, but more probably indicates contamination by a protease." @default.
