- W2051981569 abstract "1. The DNA content of isolated mitochondria from adult rat liver was determined. Evidence that the DNA measured was that of mitochondria and not due to nuclear contamination included: (1) Deoxyribose or DNA was measured by diphenylamine, cysteine, indole, tryptophan, ultraviolet absorption, and phosphate methods. All methods showed positive results, but the quantity of DNA measured by different procedures varied considerably when studied upon mitochondria. The quantity of DNA measured by these methods upon DNA standards and the “nuclear” fraction were strictly comparable. (2) Low (mean 0.65 μg DNA/mg protein) but reproducible (σ = ± 25 %) values were obtained by the diphenylamine reaction in 9 separate experiments with mitochondria from approx. 100 rat livers. (3) Feulgen-stained mitochondrial preparations showed negligible contamination by nuclei or nuclear fragments. (4) Extensive washing of mitochondrial fractions did not remove significant quantities of the diphenylamine-reactive material. (5) Addition of high polymer DNA to a mitochondrial suspension produced no increase in the DNA content of the mitochondria after they were resedimented. (6) A cell fraction of nuclei and cell debris was found to have over 90 % of its DNA digested by deoxyribonuclease (EC 3.1.4.5) in 10 min. Mitochondrial DNA was found to be only 20–30 % digestible in 4 h, even after complete disruption of the structures. 2. Evidence that a ribose-containing compound is intimately associated with the mitochondrial DNA extracts was found. The ribose-containing component, unlike the bulk of the RNA measured in mitochondria, was relatively resistant to ribonuclease (EC 2.7.7.16). Its absorption spectrum with the cysteine-H2SO4 and orcinol methods also suggests that it is a different RNA than that which is extracted by alkaline hydrolysis." @default.
