- W2052103632 abstract "RATIONALE: Previous experiments demonstrated that avoidance of peanut in peanut-allergic female mice during gestation and lactation actually enhanced peanut-hypersensitivity in their offspring. This study investigated the role of IgE in these reactions. METHODS: Intragastrically (i.g.) sensitized peanut-allergic female C3H/HeJ mice were mated 5 weeks after the initial peanut sensitization. During pregnancy and lactation, these mice were divided into 2 groups: Grp #1, no peanut exposure and Grp #2, continual low-dose peanut exposure during both gestation and lactation. Five-week-old offspring from each group were then challenged i.g with peanut. Anaphylactic reactions were scored ranging from 0, no reaction to 5, death. Peanut specific antibodies and plasma histamine levels were measured. In addition, the percent of degranulated mast cells in the ear samples was determined. RESULTS: Following challenge, anaphylactic reactions were observed in 80% of offspring in Grp #1 (total n = 10, median score 1.8, range 0-3). In contrast, only three of 10 offspring in Grp #2 showed reactions. No peanut-specific IgE was detected in mice in either group. However, significant levels of peanut specific IgG1 were present. In addition, no increase in plasma histamine levels or numbers of degranulated mast cells was found. CONCLUSIONS: The peanut-induced anaphylactic reactions in offspring of peanut allergic mice in this model are not mediated by the classic IgE/mast cell histamine release pathway. We hypothesize that the anaphylactic reactions observed are mediated by an IgG1 pathway resulting in activation of platelet activation factors." @default.
