Aus Ansätzen von 1 und Brom in Tetrachlorkohlenstoff, die zusätzlich leicht erwärmt wurden, konnte neben wenig 3 auch 1α.2-Dibrom-4.9.12β. 13.20-pentaacetoxy-2βH-6-tiglien-3-on (7) in geringen Mengen isoliert werden. Behandlung von 1 mit N-Bromsuccinimid in Tetrachlorkohlenstoff liefert im wesentlichen 3. – Behandelt man 1 mit Brom in Eisessig oder Äther/Eisessig, so wird das instabile 1.2.6.7-Tetrabromderivat 8 erhalten. Auch 8 stabilisiert sich während der chromatographischen Reinigung durch intramolekulare Acetylwanderung zum faßbaren Hauptprodukt, dem 1α.2.6-Tribrom-9-hydroxy-4.7α.12β.13.20-pentaacetoxy-2βH.6αH-tiglian-3-on (9). On the Chemistry of Phorbol, XVIII. – Bromination of Phorbol Pentaacetate By treatment of phorbol pentaacetate (1) with bromine in carbon tetrachloride at room temperature, H-10α is substituted by bromine with retention of the configuration. Concomitantly, hydrogen bromide formed in the reaction saturates Δ6.7 to yield as the primary product the labile 7β, 10-dibromo-4,9,12β,13,20-pentaacetoxy-6αH-1-tiglien-3-one (2). During chromatographic purification 2 undergoes an intramolecular acetyl migration, forming the final main product, 10-bromo-9-hydroxy-4,7α,12β,13,20-pentaacetoxy-6αH-1-tiglien-3-one (3). Gentle heating of the mixture furnished, in addition to small amounts of 3, a small quantity of 1α,2-dibromo-4,9,12β,13,20-pentaacetoxy-2βH-6-tiglien-3-one (7). By reaction of 1 with N-bromosuccinimide in carbon tetrachloride 3 is obtained as the main product. – Treatment of 1 with bromine in glacial acetic acid or ether/glacial acetic acid yields the labile 1,2,6,7-tetrabromo derivative 8. During chromatographic purification 8 also undergoes intramolecular acetyl migration to yield the final main product 1α,2,6-tribromo-9-hydroxy-4,7α,12β,13,20-penta-acetoxy-2βH,6αH-tiglian-3-one (9)." @default.
