- W2054606944 abstract "RATIONALE: Short ragweed (Ambrosia artemisiifolia) is one of the most highly allergenic pollens associated with hay fever in the U.S. Reliable methods for assessing potency of ragweed extracts are important for use in skin prick tests and immunotherapy practices. Our aim was to determine the accuracy of an Amba1 ELISA for measuring short ragweed allergen in commercial extracts compared to radial immunodiffusion (RID) and to determine cross-reactivity with other pollens. METHODS: Standardized short ragweed extracts from seven U.S. allergen manufacturers were compared for Amba1 content by polyclonal ELISA. Six ragweed species and eight standardized grass pollens were also compared for species cross-reactivity with Amba1. RESULTS: Seven standardized short ragweed extracts were labeled to contain 185-461 FDA units/mL Amba1 by RID. These extracts contained 200-387 units/mL Amba1 by ELISA. The potency differences between the two methods ranged from 4-44%. The six different species of ragweed reacted in the Amba1 ELISA, but the slopes of the standard curves were not parallel, and the ELISA could not be used for quantitative measurements in these species. The eight standardized grass pollens showed no cross-reactivity. The mean intra-assay and inter-assay variations for the Amba1 ELISA was 5.4% and 11.7% respectively. CONCLUSIONS: The Amba1 ELISA assay has consistently proven accurate as a potency assessment tool for measuring Amba1 in standardized short ragweed extracts used by U.S. allergen manufacturers. The assay compared favorably with RID and should significantly improve potency assessments and standardization of short ragweed extracts." @default.
