- W2055087123 abstract "Abstract A method has been developed with the aid of model compounds for the determination of 2-aceto-2-hydroxycarboxylic acids, 2-acetolactic acid, and 2-aceto-2-hydroxybutyric acid, in addition to vicinal diketones, in fermentation solutions. The acetohydroxy acids are separated from the diketones by running the sample through an anion-exchange column treated with bicarbonate. The column is first eluted with water. The fraction obtained contains the diketones, diacetyl, and 2,3-pentanedione, which are determined by head space gas chromatography. The acetohydroxy acids remaining in the column are eluted with a bicarbonate solution, from which they are determined as vicinal diketones by head space gas chromatography following decomposition of the acids by keeping their solution in a closed flask at 40°C for a stated time. Acetolactic acid produces diacetyl while acetohydroxybutyric acid produces 2,3-pentanedione, in amounts proportional to the concentrations of these acids in the eluate. When the method was applied to the analyses of fermentation of a solution of calcium sulfite and glucose fermented by baker's yeast, it was noticed that after 3 hr of fermentation the solution contained per liter 2.0 mg diacetyl, 2,3-pentanedione nil, 41 mg 2-acetolactic acid, and 2.5 mg 2-aceto-2-hydroxybutyric acid, and after 5 hr, at the end of fermentation, correspondingly 2.6, 0.1, 22, and 0.5 mg." @default.
