- W2055256874 abstract "Abstract From the urine of plasmocytoma patients a relatively low-molecular glycoprotein was isolated. It is heat-stable and slightly soluble in water; in trichloroacetic acid it is insoluble; on the other hand, it shows a ready solubility in 0.6 M perchloric acid. In paper and starch-gel electrophoresis at ph 7–8.5 it is uniform. Its velocity of migration corresponds to that of α 1 -globulin. The iso-electric point lies between pH 3.3 and 3.6. The sedimentation constant is 1.43. The molecular weight is 19,900 on combined measurement of the sedimentation and diffusion in the analytical ultracentrifuge. In immuno-electrophoresis it shows partial antigen community against antihuman serum in the α 1 -globulin region. It is distinguished from the total uro-protein complex and the Bence Jones protein bodies by its percentage amino acid composition and its significantly higher total carbohydrate content. It is methionine-free. The polypeptide-carbohydrate ratio is 1.5:1. The total carbohydrates make up 38.1%, which consists of 16.6% hexoses, 4.5% methyl pentoses, 10.2% hexosamines and 6.8% neuraminic acids. Hexosamine and neuraminic acid are present in the N-acetyl form. The percentage sugar distribution is Ga:Ma:Glu:Fu:Rh:G1A:GaA:NS = 8.1:4.5:4.0:3.2:1.3:8.4:1.8:6.8. The molar ratio is 10:5:5:4:2:8:2:5. The number of C-, N- and S-atoms calculated from the elementary analysis and the molecular weight found is in good correspondence with the number of C-, N- and S-atoms which can be calculated from the molar distribution. The molecular weight found being also identical to the calculated molecular weight, it may be assumed that with the 108 amino acids and 41 carbohydrates found the principal components of the glycoprotein have been demonstrated. Since this glycoprotein does not occur in urine of normal subjects and in proteinurias of different aetiology, it might be characteristic of the uroproteins in plasmocytoma." @default.
