- W2056131106 abstract "OBJECTIVE: Oxidative stress (OS) is thought to play a role in diseases of aging but changes little systemically in the decades prior to disease. The ovary ages more quickly than other organs. Markers of OS have been identified in follicular fluid, but no prior study has examined F2α-isoprostanes (IsoPs), thought to be the most reliable measure of OS status in biological specimens. We sought to determine if IsoP levels in follicular fluid correlated with markers of ovarian aging. DESIGN: A cross-sectional study involving patients undergoing ART by standard ovarian stimulation protocols. MATERIALS AND METHODS: Subjects were divided into four groups by age(<36 or >40), and oocyte number (< 25th percentile or >75th percentile). A single dominant follicle was aspirated from each of 31 patients at the time of oocyte retrieval. IsoPs were measured by stable isotope dilution GC/MS and results are shown in Table 1. These levels of OS far exceeded the relatively narrow range observed in systemic levels across this age range and were greater than the impact (p=0.07) of chronological age on FF-IsoPs. This finding suggests oxidative injury may play a critical role in the accelerated aging seen in the ovary. However, better protective mechanisms in young women vs. older women may still preserve fertility (ASRM abstract P448, 2008) This is the first study to identify IsoPs in follicular fluid and to focus on a known clinical marker of ovarian aging as the potential end result of increased OS." @default.
