• W2056247834 abstract "The EEG and behavioral features of generalized epilepsy produced in the cat by large parenteral doses of penicillin are similar to those of human ‘myoclonic petit mal’23. Earlier studies have suggested that the reduction of ascending reticular drive may be a factor in promoting generalized epileptiform discharges of this kind and that, conversely, reticular stimulation may depress or eliminate generalized epileptiform discharge altogether in this animal model.Since a cholinergic mechanism appears to be involved in mediating the cortical response to stimulation of the ascending reticular activating system, experiments were carried out using intracarotid, intravertebral and intravenous injections of drugs known to cross the blood-brain barrier which are able either to potentiate (eserine) or block (scopolamine, atropine) the action of acetylcholine.Eserine abolished or considerably reduced the epileptiform discharges for 5–60 min. With higher doses of eserine, when the bursts reappeared, they often assumed a different wave pattern and burst organization. Conversely, scopolamine and atropine were found to produce an overall increase in the amount of generalized epileptiform abnormality even though the burst organization seen prior to administration of these drugs was disrupted. These drug-induced changes (except for the late eserine effects on the reappearing epileptiform discharges) were similar to those produced by natural arousal and sleep respectively. The results provide further support for the importance of the reticular activating system as a powerful factor influencing the epileptiform discharges in this model of generalized epilepsy. The possibility that similar mechanisms may be operative in human generalized corticoreticular (‘centrencephalic’) epilepsy must be considered." @default.
