- W2056480244 abstract "Abstract A mathematical model was developed which permitted one to determine the total affinity constant ( K t ) pf heterogenesous hapten-antibody interactions. The total affinity constant ( K t ) represents the sum of the weighted affinities of the subpopulations in heterogeneous hapten-antibody interactions. This was verified by using experimental data on the binding of ϵ-dinotrophenyl- L -lysine with rabbit anti-dinitrophenyl Immunoglobulin G (IgG) and bovine anti-dinitrophenyl Immunoglobulin M (IgM). Computer least squares calculated values for the association constants ( K ′s) and the concentrations of antibody binding sites ( N ′s) for various assumed subpopulations were obtained by the use of an iterative and steepest descent non-linear computer program to best fit the experimental data points of R as a function of C . These calculated parameters and ( K and N ) were then used to calculate the total affinity constant ( K t ) by the use of the mathematical model derived in this investigation. The K t 's could also be obtained by a graphical procedure and were found to be in excellent agreement with the computer calculated values." @default.
