- W2056659547 abstract "Abstract The process of double-stranded RNA bacteriophage φ6 penetration was studied by examining the parental viral proteins and RNA after infection. A host bacterium, Pseudomonas phaseolicola , was infected with phage φ6 labeled with [ 3 H]uracil and [ 35 S]methionine. The infected cells were subjected to blender and subsequent lysozyme-EDTA treatments. After these treatments, substantial amounts of both 3 H and 35 S radioactivity were retained in the cellular fraction, and the ratio of 35 S/ 3 H was little reduced. This indicates that the majority of the phage proteins penetrated the host cell surface with the genome RNA. Most of the cell-associated radioactivity after the blender treatment was released by ultrasonic cell disruption and it was analyzed by sucrose gradient centrifugation. Radioactivities of both parental proteins and RNA were separated into three fractions, I, II, and III, in order of sedimentation rate. Fraction I was shown to be a subviral particle fraction containing intact RNA and four species of proteins (P1, P2, P4, and P7). Fraction I also contained viral RNA newly synthesized in the infected cell. This indicates that Fraction I particle is the site of the initial viral RNA synthesis." @default.
