- W2056797795 abstract "Treatment of Escherichia coli ribosomes (70-S) in 5 mM Mg2+-50 mM KCl-30 mM Tris-HCl buffer (pH 7.4) with nitrogen mustard resulted in dissociation of these particles into 30-S and 50-S subunits. This dissociation reaction was dependent upon the concentration of the reagent, and the time and temperature of incubation. The ionic environment (Mg2+, KCl) also played an important role: increasing the concentration of Mg2+ resulted in decreased dissociation, whereas increasing the concentration of KCl resulted in increased dissociation. Mn2+ and Ca2+, and spermidine and spermine, but not putrescine and cadaverine, exhibited a protective effect similar to that of Mg2+. The addition of these divalent cations or polyamines to ribosome preparations which had been treated with nitrogen mustard resulted in essentially complete reversal of ribosomal dissociation. The 70-S particles thus re-formed were active in phenylalanine incorporation directed by poly (U) in a cell-free system. Possible mechanisms involved in this reversible dissociation are discussed." @default.
