- W2057295181 abstract "From a rat-liver homogenate three fractions containing free polyribosomes, rough membranes and smooth membranes, respectively, were isolated in high sucrose concentrations. The effects of these fractions on the incorporation of [14C]leucine in a cell-free system were as follows: 1. In the presence of free polysomes, the incorporation of [14C]leucine was stimulated by the addition of non-labeled amino acids. 2. When rough membranes were suspended in a medium with low sucrose concentrations (< 1 M) the suspension became turbid. The addition of the non-labeled amino acids then had no stimulatory effect on the incorporation of [14C]leucine. However, when rough membranes were suspended in high sucrose concentrations (> 1 M) or in postmicrosomal supernatant fraction, the solution remained clear and the addition of non-labeled amino acids caused a stimulation of [14C]leucine incorporation. 3. Freshly isolated smooth membranes mixed with free polyribosomes behaved like rough membranes with respect to amino acid incorporation. The loss of the stimulatory effect of the cold amino acids on [14C]leucine incorporation, after the mixture was suspended in a medium without sucrose, was due to the membranous part. 4. When isolated rough membranes were resubjected to the procedure used for the fractionation of total rat-liver homogenate, again a fraction with free poly-ribosomes was obtained, provided that the rough membranes were suspended in a medium with high sucrose concentrations. 5. Polyribosomes isolated by treatment of rough membranes with deoxycholate contained 15 % vacant ribosomes, i.e. ribosomes not attached to mRNA. In this respect they were similar to free polyribosomes since these also contained approx. 15 % vacant ribosomes." @default.
