L’hyperglycémie, caractéristique du diabète, joue un rôle important dans la survenue des complications diabétiques en favorisant la glycation des protéines et l’accumulation de produits terminaux de glycation (PTG). Les PTG sont un ensemble hétérogène de composés résultant de réactions non enzymatiques (réaction de glycation ou réaction de Maillard) entre des sucres réducteurs et des groupements amines de molécules biologiques comme les protéines, les lipides ou les acides nucléiques. Chez des patients diabétiques, une élévation des taux sériques de PTG est corrélée avec la présence de complications. Le rôle des PTG dans les néphropathies, les neuropathies, les rétinopathies et les complications cardiovasculaires au cours du diabète a fait l’objet de nombreux travaux au cours de ces 20 dernières années. Un intérêt grandissant porte actuellement sur les composés inhibiteurs de la glycation du fait de leur potentiel préventif ou thérapeutique. Hyperglycemia, the hallmark of diabetes, plays an important role in the onset of complications in diabetic patients by promoting protein glycation and accumulation of advanced glycation end products (AGEs). The AGEs belong to a heterogeneous group of compounds resulting from non-enzymatic reactions (Maillard reaction or glycation) between sugars and amino groups of biological molecules such as proteins, lipids or nucleic acids. In patients with diabetes, elevated serum levels of AGEs are correlated with the presence of complications. The role of AGEs in nephropathy, neuropathy, retinopathy and cardiovascular diseases in diabetic patients has been the subject of numerous studies over the past 20 years. A growing interest is currently focused on compounds that inhibit glycation because of their preventive or therapeutic potential." @default.
