- W2057483764 abstract "Abstract Equine abortion virus was purified from the plasma of infected golden Syrian hamsters by differential centrifugation and nuclease treatment. The preparations were essentially free of nonviral elements on electron microscopic examination, and sedimentation in sucrose and potassium tartrate density gradients resulted in a single visible band. Electron microscopy of this band showed it to be composed of viral particles, and injection into hamsters resulted in infection and death of the animals. The viral particles had a sedimentation coefficient of approximately 2200 S and a hydrated density of 1.18. Measurement of particle diameter from shadowed preparations in the electron microscope gave an average diameter of 167 mμ; however, calculation of the particle diameter from sedimentation and density data resulted in an average diameter of 148 mμ. On the basis of particle counts and colorimetric determinations, there was an average of 4.17 × 10 −10 μg of DNA per particle. This comprised about 9.2% of the 4.53 × 10 −9 μg estimated dry weight of the particle. Chromatograms of hydrolyzed viral preparations showed only the major constitutive bases of DNA. The bases were paired, adenine approximately equal to thymine and guanine to cytosine. The molar dissymmetry ratio (A + T/G + C) of the viral DNA was 0.78 in contrast to the ratio of 1.40 for host cell DNA." @default.
