- W2057982616 abstract "In order to investigate the effect of α,β-unsaturated aldehydes on tissue preservation prepolymerized glutaraldehyde or glutaraldehyde-formaldehyde agents were prepared. The polymeric aldehydes were made up from glutaraldehyde alone or from mixtures of glutaraldehyde and formaldehyde. The aldehydes were heated and then freeze-dried or distilled in vacuo. The chemical nature of the aldehyde preparations was investigated by u.v. spectroscopy, i.r. spectroscopy, elemental analysis and [13C]NMR spectroscopy. The results of the analyses indicate that the aldehyde preparations contain a high amount of unsaturated oligomers. The effect of the prepolymerized aldehydes were tested by electron microscopy. Immersion fixation of liver tissue showed that the prepolymerized aldehyde preparations only provided an acceptable fixation in the most superficial part of the tissue. It is suggested that the surface fixation caused by the prepolymerized aldehydes is so effective that it hinders the fixative solutions to penetrate deeper into the tissue. Thus, fixation might first be established when mono- or dimeric species have penetrated into the tissue where they are responsible for the cross-binding reactions to take place." @default.
