- W2058147983 abstract "Abstract Two-cell mouse embryos were flushed from the oviducts 40 hours after HCG-injection of female CB6F1 mice. They were cultured for 72 hours from two-cell to blastocyst stage in two different dioxin-contaminated culture media. During this time the embryo development was visually monitored. The culture media used were M 16 medium, supplemented with 0.4% BSA, and 2nd trimester human amniotic fluid. The dioxin concentrations in amniotic fluid were 7.4 ng/ml for 1,2,3,4,7,8-HexaCDD and 5.5 ng/ml for 1,2,3,4-TetraCDD. No dioxins could be detected in control amniotic fluid. The achievable dioxin concentrations in M 16 medium were 55 ng/ml for 1,2,3,4-TetraCDD, 1.9 ng/ml for 1,2,3,7,8-PentaCDD, 5.6 ng/ml for 1,2,3,4,7,8-HexaCDD and 7.8 ng/ml for 1,2,3,4,6,7,8,9-OctaCDD. For each congener in M16 medium 170 embryos were used to test for the influence on preimplantation development. While in control medium only 18.1% of all embryos tested showed signs of degeneration, the degeneration rate in 1,2,3,4-TetraCDD-contaminated medium was 50.9% and in 1,2,3,4,7,8-HexaCDD-contaminated medium 50.3% after 72 hours in culture. No effects could be detected in OctaCDD-contaminated medium. For each congener in amniotic fluid 110 embryos were cultured. The degeneration rate for control medium was 6.5%, for 1,2,3,4-TetraCDD 20% and 22.3% for 1,2,3,4,7,8-HexaCDD. We conclude that dioxins interfere with the preimplantation embryo development in low concentrations. The negative dioxin influence on embryonic development was diminished in human amniotic fluid as compared to synthetic M 16 medium." @default.
