- W2058358712 abstract "A transplantable pituitary tumor induced in a male C57Bl mouse by radiothyroidectomy has been studied by light and electron microscopy. The primary tumor was highly differentiated and composed of hypersecretory cells which showed the morphological characteristics of the thyroidectomy cell. The tumor was followed through four transfers in radiothyroidectomized hosts. Autonomous tumors growing in normal hosts appeared during the fifth transfer generation. The first dependent tumor grafts showed highly differentiated cells of the same appearance as the primary tumor cells. Through the following transfers there was a progressive dedifferentiation of the tumor cells although the majority of them could, in the electron microscope, be identified as thyrotrophic tumor cells with morphological signs of secretory function. In the autonomous tumors most of the cells were dedifferentiated and only a few cells showed signs of secretory function. The results of the morphological study support earlier observations on the hormone activity of transplantable thyrotrophic tumors. These have shown that primary and dependent tumors are highly functional, but that autonomous tumors display a progressive decrease in hormone production. The findings also confirm earlier morphological observations that autonomous tumors are dedifferentiated, but both the light and electron microscopic observations in this study suggest that there is a progressive dedifferentiation of tumor cells in hormonedependent tumors as well. Étude en Microscopie Optique et Électronique D'une Tumeur Transplantable de L'hypophyse (Zone Thyréotrope) Chez la Souris On a étudié en microscopie optique et électronique une tumeur hypophysaire transplantable induite par radiothyroïdectomie chez une souris mǎle C57Bl. La tumeur primitive était hautement différenciée, se composant de cellules hypersécrétantes qui présentaient les caractères morphologiques de la cellule après thyroïdectomie. On a suivi la tumeur pendant quatre transplantations sur des hǒtes radiothyroïdectomisés. Des tumeurs autonomes se développant chez l'hǒte normal sont apparues pendant la cinquième génération de transferts. Les premières greffes de tumeurs dépendantes montraient des cellules hautement différenciées, ayant le měme aspect que les cellules tumorales primitives. Pendant les transplantations suivantes, on a assisté à une dédifférenciation progressive des cellules tumorales; toutefois, au microscope électronique, on pouvait reconnaǐtre dans la majorité d'entre elles des cellules tumorales thyréotropes présentant des signes morphologiques de fonction sécrétoire. Dans les tumeurs autonomes, la plupart des cellules étaient dédifférenciées et seules quelques-unes présentaient des signes de fonction sécrétoire. Les résultats de l'etude morphologique viennent renforcer les observations antérieures sur l'activité hormonale des tumeurs thyréotropes transplantables. Celles-ci montraient que les tumeurs primitives, dépendantes, sont hautement fonctionnelles tandis que les tumeurs autonomes accusent une diminution progressive de la production hormonale. En outre se trouvent confirmées les observations morphologiques montrant que les tumeurs autonomes sont dédifférenciées mais, dans la présente étude, les observations tant au microscope optique qu'au microscope électronique font penser qu'il y a aussi une dédifférenciation progressive des cellules tumorales dans les tumeurs hormonodépendantes." @default.
