The components of tissue extracts are separated by cation chromatography and analyzed both fluorometrically by the orthophthaldialdehyde and mercaptoethanol reaction, and by scintillation spectrometry. The sensitivity of fluorometric detection was 40 pmol/sample. The method was applied to the measurement of the specific radioactivity of dopamine in corpus striatum synaptosomes following incubation in Krebs-bicarbonate medium containing isotopically labelled DOPA. The specific radioactivity of dopamine increased linearly with the time of incubation in the presence of the isotope. Incubation in the presence of 56 mm potassium caused a significant increase in the total dopamine (nmol/100 mg protein) which was reflected by a decrease in the specific radioactivity of dopamine." @default.
