- W2059105887 abstract "This chapter provides an understanding of how a buffer will affect Ca2+ homeostasis within a cell and discusses the relevant parameters that need to be considered. Most studies dealing with EF-hand Ca2+ binding proteins focus either on their metal-binding affinities (KD values) or elucidate their intracellular localization within specific cell types in a given tissue. Two types of Ca2+ binding sites in EF-hand proteins have been identified on the basis of differences in their selectivity and affinity for Ca2+ and Mg2+ ions. The role played by Ca2+ buffers is apparently a simple one; namely, to bind this cation and thereby lower [Ca2+]. However, soluble buffers represent but one component of the intricate system implicated in Ca2+ homeostasis. The distinct properties of each buffer are also illustrated by the “compensation” or “homeostatic” mechanisms that are induced in their absence in knockout mice. Ca2+ buffers thus constitute an integral part of the finely tuned system involved in Ca2+ homeostasis, and have a profound effect on many aspects of Ca2+ signaling. The removal of such a buffer does not apparently trigger the obvious compensation mechanism (that is, the upregulation of another Ca2+ buffer), but leads rather to subtle changes in cell morphology or to discrete modulations in Ca2+ uptake or release systems.
