- W2059230873 abstract "The solvent perturbation technique of difference spectroscopy has been used to study the interaction of an anti-synthetic polypeptide antibody with its homologous antigen. Antibody was obtained from a sheep which had been immunized with the random polymer (Glu 60 Ala 40 ) n (GA). This particular preparation, isolated by either immunoabsorbent columns or dissolution of a specific precipitate, requires divalent cations (especially calcium) to affect interaction with the antigen (Maurer et al. , 1970; Liberti et al. , 1971; Callahan et al. , 1974. Perturbation differences spectra were recorded in 4 different solvents (dimethyl sulfoxide, ethylene glycol, glycerol and polyethylene glycol) at wavelenghts between 310 and 270 nm. Since the antigen is transparent in this region, all absorbance differences could be attributed to the immunoglobulin molecule. Antigen-antibody solutions (at ∼ 20-fold equivalence excess of GA) were studied in both the absence and presence of Ca ++ and chromophore exposure was calculated in each case. With the smaller perturbants, approximately 8–10 tryptophane and 31–38 tyrosine residues appear exposed to solvent prior to the addition of calcium. The larger perturbant (PEG) affects ∼ 5 tryptophane and 28 tyrosine residues. Subsequent to calcium addition, three tyrosine residues per antibody combining site become unavailable to smaller perturbants. With polyethylene glycol, however, only one tyrosine is affected. These data are interpreted to mean that each combining site contains three tyrosine residues which are involved in binding antigen and that two of the three may lie in a ‘crevice’ or fold in the protein surface." @default.
