- W2059282978 abstract "Polymorphism in the assembly of sickle cell hemoglobin has been studied by electron microscopy of negatively stained fibers of hemoglobin S obtained by lysis of sickled cells with phosphotungstic acid. The structures observed include linear strands of hemoglobin molecules (designated monofilaments), helical fibers composed of multiple monofilaments and larger associations of monofilaments into multistrand cables and two-dimensional sheets. A survey of several thousand particles showed that at least two distinct species of helical fibers can be found in sickle cell lysates. The one more commonly found in the study is a helical fiber often up to one micrometre in length and with a diameter in the range of 180 to 220 Å. Optical transforms of these fibers display meridional diffraction maxima corresponding to a spacing along the fiber axis of 29 Å and off-meridional maxima corresponding to a spacing along the fiber axis of 57 Å. In addition, near-equatorial maxima are observed and these correspond to a variable spacing along the fiber axis of 300 to 550 Å. The orders of these diffraction maxima show that the fibers are composed of eight strands of monofilaments arranged in helical tubes with each strand staggered with respect to the neighboring strand by half of a hemoglobin molecule. Variations in the pitch of the monofilaments account for the presence and position of the near-equatorial diffraction maxima. The structure also can be described as a four-start helix with each start defined by the loci of pairs of adjacent monofilaments. The second form of helical fiber observed in cell lysates is a six-stranded stacked disc structure identical to that described by Finch et al. (1973). Optical transforms of hemoglobin S fibers inside sickle cells are also presented and in these diffraction maxima at 29 Å and 57 Å occur not as discrete spots but as arcs, reminiscent of X-ray diffraction patterns of fibers with the arcing due to the lack of perfect orientation among the fibers in the array." @default.
