- W2059652075 abstract "395 There is evidence that cell bioenergetics are altered by cell O2 tensions, such that at similar rates of cell respiration and work output, there is a greater disruption of cell homeostasis with lower PO2. The purpose of this study was to examine the relationship between muscle PCr hydrolysis and the development of fatigue in human calf muscle, while the subjects breathed varied inspired O2 fractions (FIO2). Six subjects performed plantar flexion exercise on three separate occasions. Work intensity was incrementally increased (1 watt increments starting at 0 watts) every 2 min until exhaustion was attained. The only variable altered for the exercise bouts was the FIO2 breathed (in a blocked order design so that all six possible orders of the 3 gases were used): being either 0.1, 0.21, or 1.00 O2 in N2. Muscle PCr hydrolysis was monitored every 4 sec by 31P-MRS. Time to exhaustion (and thereby work intensity achieved) was significantly (p<0.05) greater as FIO2 was increased (17.7± 1.3 min, 21 ± 0.9, and 24 ± 1.8 during 0.1, 0.21, and 1.00 O2 breathing, respectively). Even with the significantly greater work intensities achieved as FIO2 was increased, muscle [PCr] at exhaustion was not significantly different among the three treatments(41%±3, 44%±3, and 43%±4 of resting [PCr]). Muscle [PCr] was significantly more depleted from the 5 watt work intensity until exhaustion in the FIO2 =0.1 condition compared to the other two FIO2 conditions. [PCr] degradation was never significantly different at any work intensity between the FIO2=0.21 and 1.00. These results demonstrate that even at equivalent submaximal work intensities (and certainly equivalent rates of respiration), PCr hydolysis is greater with reduced O2 tension. It is likely that this greater disruption of homeostasis while breathing a low FIO2 contributes to the early onset of fatigue." @default.
