- W2061743281 abstract "We and others have demonstrated that androgenic compounds are able to reduce cervical resistance (induce cervical ripening). The mechanism responsible for this effect has yet to be determined. We hypothesize that androgens directly induce ripening through androgen receptors. The purpose of this study was to investigate the effect of androgen receptor inhibitor Flutamide on rat cervical ripening and proteoglycan remodeling. Timed pregnant Sprague-Dawley rats were treated with 10 mg of Flutamide beginning on day 12 of gestation. Animals were gavage fed with vehicle or Flutamide daily through day 20. We assessed cervical resistance by cervical creep on day 21. Glycosaminoglycans (GAGs) composition and chain length was assessed by Fluorophore-assisted carbohydrate electrophoresis (FACE). Proteoglycan core protein mRNA expression was determined by real-time RT-PCR. Flutamide inhibited the gradual decline in cervical tensile strength that occurs during the second half of the pregnancy. Flutamide induced a decrease in hyaluronan content, an increase in disulfated disaccharide content, and a shortening in the chain length of chondroitin/dermatan sulfate. Additionally, Flutamide up-regulated aggrecan mRNA and tend to down-regulate decorin and versican mRNA and up-regulate fibromodulin mRNA. Flutamide produced no changes in the content of non-sulfated, 4-sulfated or 6-sulfated disaccharide. Flutamide-inhibited rat cervical ripening is associated with changes in GAG composition and chain length. The gradual decline in cervical resistance during the second half of gestation may be due to increased androgenic steroids that up-regulate GAG content and modulate Proteoglycan remodeling." @default.
